A preclinical evaluation of an autologous living hyaline-like cartilaginous graft for articular cartilage repair: a pilot study
نویسندگان
چکیده
In this pilot study, an autologous synthetic scaffold-free construct with hyaline quality, termed living hyaline cartilaginous graft (LhCG), was applied for treating cartilage lesions. Implantation of autologous LhCG was done at load-bearing regions of the knees in skeletally mature mini-pigs for 6 months. Over the course of this study, significant radiographical improvement in LhCG treated sites was observed via magnetic resonance imaging. Furthermore, macroscopic repair was effected by LhCG at endpoint. Microscopic inspection revealed that LhCG engraftment restored cartilage thickness, promoted integration with surrounding native cartilage, produced abundant cartilage-specific matrix molecules, and re-established an intact superficial tangential zone. Importantly, the repair efficacy of LhCG was quantitatively shown to be comparable to native, unaffected cartilage in terms of biochemical composition and biomechanical properties. There were no complications related to the donor site of cartilage biopsy. Collectively, these results imply that LhCG engraftment may be a viable approach for articular cartilage repair.

Articular cartilage covers the end of bones in joints and determines the load-bearing characteristics and mobility of joints. It has a thin, smooth, low friction surface with a remarkable resiliency to compressive forces. In general, chondrocytes occupy lacunae in the matrix, and produce cartilaginous ECM (extracellular matrix), which consists of type II collagen (13%), proteoglycans (7%), and ...
متن کاملMinced articular cartilage--basic science, surgical technique, and clinical application.
Minced articular cartilage procedures are attractive surgical approaches for repairing articular cartilage, as they are 1-staged, autologous, and inserted on a carrier that can potentially be placed arthroscopically. The principle of mincing the autologous donor cartilage is to create a larger surface area for cartilage expansion. Placement on a scaffold carrier allows for a chondro-inductive a...

The extracellular matrix (ECM), present between cells, is the authentic substrate for most cells in living organisms. Interplay between the ECM and cells plays a critical role in regulating cell differentiation, proliferation, apoptosis, and matrix synthesis. A substantial characteristic of hyaline cartilage is that a few chondrocytes, which are the sole cells in this tissue, are surrounded by ...

OBJECTIVE Cartilage-derived morphogenetic protein 1 (CDMP1), which is a member of the transforming growth factor-beta superfamily, is an essential molecule for the aggregation of mesenchymal cells and acceleration of chondrocyte differentiation. In this study, we investigated whether CDMP1-transfected autologous bone marrow-derived mesenchymal cells (BMMCs) enhance in vivo cartilage repair in a...

Tissue engineering is an increasingly popular method of addressing pathological disorders of cartilage. Recent studies have demonstrated its clinical efficacy, but there is little information on the structural organisation and biochemical composition of the repair tissue and its relation to the adjacent normal tissue. We therefore analysed by polarised light microscopy and immunohistochemistry ...
